The Birth of Synthetic Biology: From Reading Life to Writing Life
> In 1978, scientists at Genentech celebrated a milestone in their laboratory: they successfully engineered *Escherichia coli* to produce human insulin. This was the first time in human history that a gene from another organism was transplanted into *E. coli* to enable it to produce a protein that could previously only be manufactured by humans. Just 25 years had passed since the discovery of the DNA double helix structure, and more than 20 years remained before humans would first sequence a complete genome. This event is regarded as the earliest "prototype" of synthetic biology, marking humanity's official transition from the era of "reading life" to the threshold of "attempting to write life".

> In 1953, James Watson and Francis Crick of the University of Cambridge published a two-page paper in *Nature* announcing the DNA double helix structural model. This model revealed the core storage rules of genetic information: genetic information is stored in the sequence of four bases (adenine A, thymine T, guanine G, cytosine C) on the double strand of DNA, with the two strands connected via complementary base pairing (A pairs with T, G pairs with C). This pairing rule ensures that genetic information can be accurately replicated during cell division, and also determines that genetic information can be transferred from DNA to RNA, and then to proteins, completing all steps of the "central dogma" of molecular biology.
> If the double helix structure is the "grammar book" of life sciences, then sequencing technology is the tool humans use to "read" the source code of life. In 1977, British biochemist Frederick Sanger invented the chain termination sequencing method (Sanger sequencing), the first technology in human history that could accurately determine DNA sequences, for which Sanger won the Nobel Prize in Chemistry twice. The principle of Sanger sequencing uses fluorescently labeled dideoxynucleotides (ddNTPs) to terminate the extension of DNA chains, and separates DNA fragments of different lengths via capillary electrophoresis to read the corresponding base sequence. This technology remains the gold standard for small-fragment DNA sequencing to this day.
> The advent of Sanger sequencing
The successful completion of the HGP marked the official arrival of the "reading life" era. According to statistics from the National Human Genome Research Institute (NHGRI), the total cost of the first human genome sequencing was about $2.7 billion, and it took 13 years to complete; by 2023, the cost of sequencing a complete human genome has dropped to less than $200, and the time required is only a few hours. This 10,000-fold drop in cost and 100,000-fold increase in speed is the core driving force for the transformation of life sciences from "qualitative research" to "quantitative research", and also laid the data foundation for the birth of synthetic biology. The HGP not only provided the complete "source code" of human life, but also promoted the development of a large number of related technologies: bioinformatics analysis tools, high-throughput sequencing technology, gene synthesis technology, etc., all of which sprouted and developed during the HGP.
> 💡 电子版侧边栏:人类基因组计划中,中国承担了1%的测序任务,即人类3号染色体短臂上约3000万个碱基对的测序工作,这是中国第一次参与 such a large-scale international scientific project, also laid the foundation for China's subsequent leading position in the field of synthetic biology.
After the HGP, the rise of next-generation sequencing (NGS) technology further lowered the threshold of gene sequencing. Unlike Sanger sequencing which can only sequence one DNA fragment at a time, NGS technology can sequence millions to billions of DNA fragments in parallel at one time, which greatly improved the sequencing efficiency. In 2005, the first commercial NGS platform was launched, and by 2022, the global NGS market size has exceeded $25 billion. The wide application of NGS technology makes it possible to sequence the entire genome of any organism at an ultra-low cost, which also provides a powerful tool for the design and construction of synthetic biological systems.
Although the "reading life" era has enabled humans to master the genetic source code of almost all known organisms, simply "reading" cannot solve many major challenges facing mankind: such as the global food crisis, the shortage of energy and resources, the prevalence of incurable diseases, and environmental pollution. Traditional molecular biology and genetic engineering rely on random screening and modification of naturally existing genes, which is inefficient and difficult to achieve complex functions. For example, traditional metabolic engineering can only modify one or a few genes in the existing metabolic pathway of microorganisms, and it is difficult to construct a brand new metabolic pathway to produce compounds that do not exist in nature. This limitation has prompted scientists to think: can we not just "read" the genetic code, but also "write" the genetic code according to our own needs, design and construct biological systems with preset functions?

The transformation from "reading life" to "writing life" did not happen overnight, but went through more than 20 years of technical accumulation after the completion of the HGP. The key to this transformation is the breakthrough of DNA chemical synthesis technology and genome transplantation technology.
In 2000, a team led by Hamilton Smith, a Nobel laureate in Physiology or Medicine, successfully synthesized the first complete artificial genome in human history: the genome of φX174 bacteriophage, with a total length of 5386 base pairs. This genome was synthesized by chemical synthesis of short DNA fragments, followed by assembly in vitro, and then introduced into *Escherichia coli*, where it can complete the entire life cycle of the phage like a natural phage.
