Chapter 1: The Millennium-long Dream of Rewriting Life: From Myths and Legends to the Germination of Early Gene Editing Technologies
Dating back to the late Upper Paleolithic period roughly 30,000 years ago, the walls of the Lascaux Cave in France bear paintings of running bison and antlered deer. These murals are not just the dawn of art, but a metaphor for humanity’s earliest attempt to "record and reshape life": early humans longed to understand the habits of animals to domesticate them for their own use; they hoped to conquer disease, extend lifespan, and even control their own destiny. This obsession, embedded in the very genes of human civilization, has traversed myths, agrarian revolutions, and scientific revolutions, ultimately coalescing into the technology of "gene editing" that will reshape human destiny in the 21st century.
This obsession first crystallized in human collective memory in the form of myths. In the Chinese mythological system, the legend of Nüwa "molding humans from clay" is not a figment of imagination: the clay figures she crafted came to life, breathed, walked and reproduced, which is essentially humanity’s earliest imagination of "creating new life forms". A parallel narrative exists in Western mythology: Prometheus, the Titan, shaped humans in the image of the gods from clay, and Athena breathed a soul into them to give them life. The human desire to "transcend natural limits and rewrite the trajectory of life" took root from these legends. A more explicit expression of this obsession is ancient emperors’ pursuit of immortality: Emperor Qin Shi Huang sent Xu Fu, with 3,000 virgin boys and girls, eastward to seek the elixir of life. Xu Fu’s expedition eventually settled in Japan, and the "elixir of immortality" he sought was essentially humanity’s earliest practice of trying to rewrite the limits of its own life through external substances — only at that time, humans did not yet know that the code of life was hidden in the nucleus of every cell.
Beyond myths, early human practices were unconscious "gene selection". About 10,000 years ago in the Neolithic Age, inhabitants of the Fertile Crescent began domesticating wild wheat: they selected plants with larger grains and shorter stalks to留种留种 each year, and after more than a thousand generations of selection, they finally obtained the wheat varieties we eat today. The "regional planting method" recorded in the Han Dynasty agricultural book *Fan Shengzhi Shu* requires farmers to select plump seeds that are "yellow and glossy, without insect damage" for separate planting, which is essentially a directional change in crop traits through artificial selection — only at that time, humans did not know that the "large grain" and "pest resistance" traits they selected were determined by specific genes in the crop genome. This unconscious gene selection was even specifically mentioned by Darwin in *On the Origin of Species*: he counted more than 150 varieties of pigeons, from the fantail, pouter, and carrier pigeon domesticated from the rock dove, whose trait differences are so large that they look like different species. The core of domestication is human directional selection of beneficial variations. The efficiency of this selection was extremely low: it often took decades or even hundreds of years to obtain a crop variety with stable improved traits, and the human desire for "rapidly rewriting life" never stopped.
Human systematic exploration of the code of life began with the genetic revolution of the 19th century. In 1856, Gregor Mendel, a monk at the Augustinian Abbey in Brno, Austro-Hungarian Empire, began his pea hybridization experiment. The reason he chose peas was that peas are self-pollinating, cleistogamous plants, naturally purebred, and have 7 pairs of stable, easily distinguishable relative traits: tall and short stems, round and wrinkled grains, yellow and green cotyledons... He planted 7,324 pea plants in the abbey’s garden and counted them continuously for 8 years, finally finding that the separation ratio of dominant and recessive traits in hybrid offspring was stably 3:1. He summarized this as the "law of segregation of hereditary factors" and the "law of independent assortment", and published the epoch-making paper "Experiments on Plant Hybridization" in 1865. Mendel’s conclusion completely negated the then-popular "blending inheritance" theory — the latter believed that parents’ traits would be passed on to offspring like paint mixing, for example, a black person and a white person would have a "gray-skinned" child. Mendel proved that inheritance is discrete and quantifiable: each trait is controlled by two independent "hereditary factors" (later named "genes"), one from each parent, which separate from each other when forming gametes.
However, Mendel’s discovery was not immediately recognized. His paper was buried for 35 years, until 1900, when three scientists — Hugo de Vries of the Netherlands, Carl Correns of Germany, and Erich von Tschermak of Austria — rediscovered Mendel’s laws through their own experiments respectively, and genetics officially became an independent discipline. In 1909, Danish scientist Wilhelm Johannsen first proposed the term "gene" to refer to the "hereditary factor" mentioned by Mendel. At this time, humans still did not know what the chemical nature of the gene was, until 1944, when the experiment by Oswald Avery, Colin MacLeod, and Maclyn McCarty proved that the substance causing bacterial transformation was DNA, not protein, and humans realized for the first time that DNA is the carrier of genetic information.
In 1953, the era of molecular biology officially arrived. Young scientists James Watson and Francis Crick at the Cavendish Laboratory of Cambridge University, inspired by the X-ray diffraction photo of DNA (the famous "Photo 51") taken by physicist Rosalind Franklin and her student Raymond Gosling, combined with the base pairing rules discovered by biochemist Erwin Chargaff (A pairs with T, G pairs with C, i.e., $A=T, G=C$), constructed the model of the DNA double helix structure. The core significance of this structure
